By using custom columns with CPGs preloaded with nucleosides, we’re able to cleave our oligos under extremely mild conditions, preserving the 3’-end of our long sequences,” they shared. “Under the same condition using standard columns with Unylinker chemistry, we have a Unylinker adduct on the 3′-end of the oligo. In order to remove that, we do need to heat up the cleaved oligo to 65C, but then our long oligo is also damaged along the way.

Since our oligo is really long, we know for a fact that we want to use CPG with 2000A pore size, which is not an optionality offered with standard off-the-shelf synthesis columns. Assuming the answer to all 3 is yes, then I try their products a few times. Do they make custom column of a high quality – say, consistent loading, and consistent flow characteristics? Etc. (For the record – you and your applications team score A+ through and through.)
